Biografia Figlio d'arte, nacque da Zachary Nugent Brooke (1883–1946), membro della British Academy e medievista a Cambridge, e da sua moglie Rosa Grace Brooke (1888–1964). Dopo aver frequentato il Winchester College, intraprese gli studi universitari al Gonville and Caius College di Cambridge, dove fu allievo di David Knowles.
Brooke trascorse i suoi primi anni come capo dipartimento al Westfield College dell'Università di Londra, prima di assumere un incarico al Caius dal 1977 al 1994, dove rimase membro a vita. Ricoprì la carica di professore di Storia della Chiesa a Cambridge prima di diventare emerito. Fu presidente della Ecclesiastical History Society (dal 1968 al 1969).
Brooke conobbe la sua futura moglie e collega medievista Rosalind Brooke (nata Clark) a Cambridge nel 1951. Lei morì nel 2014.
Brooke si spense il 27 dicembre 2015, all'età di 88 anni.
